The CAIX inhibitor S4 is a potent and selective inhibitor of carbonic anhydrase IX/XII (CA IX/XII) with Ki of 7 nM and 2 nM, respectively. The CAIX inhibitor S4 also inhibited CA II and CA I (Ki of 546 and 5600 nM, respectively). The CAIX inhibitor S4 can suppress the number of lung metastases in an orthotopic MDA-MB-231 mouse model without affecting primary tumor growth.
